Methodology

To fulfill the above-mentioned objectives we will use the following methodology and approaches:
1)Standard techniques used to grow and manipulate fugal and bacterial cells.
2) Standard transformation methods for Saccharomyces cerevisiae and Escherichia coli cells.
3)Standard molecular biology techniques (nucleic acid isolation/purification/quantitation, Northern, Western blotting, primers design, PCR gene analysis, confocal microscopy, cell imaging, RT-PCR.
4)Gene disruption.
5)Phase contrast and fluorescence microscipy techniques.
6)Bioluminiscence assays.
7)Reporter gene assays (beta-galactosidase assay).
8)Isolation and purification of plant antioxidants.
9)Atomic absorption spectroscopy, UV-Vis spectroscopy.
10)Mass spectrometry (GC/MS).
11)Kinetic studies of metal transport within cells.
12)Etc.
