Aspecte teoretice si practice ale culturilor de celule animale

 

Cursul (28 h) se adreseaza studentilor din anul I de studiu din cadrul Masterului de Biochimie si Biologie Moleculara si abordeaza urmatoarele subiecte: 

1. Istoric, obiective, avantajele si dezavantajele culturilor celulare;

2. Sisteme de cultivare a celulelor animale: Culturile tisulara, de organ si de celule; Culturile organotipica si histotipica; Culturi la densitati celulare mari si clonale; Culturi de celule aderente si in suspensie; Culturi primare, linii celulare finite si continue;

3. Medii de cultura si conditii de cultivare; 

4. Ciclul de creştere si proliferarea celulelor in cultura

5. Sincronizarea celulara: Metode chimice; Metode fizice;

6. Imortalizarea celulara;

7. Transfectia celulara: Tipuri de transfectie; Conditii specifice de cultivare a celulelor; Metode de transfectie celulara;

8. Transformarea celulara: Instabilitatea genomica; Imortalitatea celulara; Controlul aberant al cresterii celulare; Tumorigenicitatea celulelor transformate;

9. Senescenta celulara: Tipuri de senescenta si caracteristicile celulelor senescente; Cai moleculare de activare a senescentei celulare; Mecanisme de inducere a senescentei;

10. Latenta celulara, diferentierea si de-diferentierea celulara;

11. Citometria in flux;

12.  Aplicatii ale culturilor celulare in obtinerea anticorpilor monoclonali.
